Our goal is to understand in quantitative terms the basic mechanisms involved in the repair of DNA break by homologous recombination (HR), their interplay with the higher "cytological" order of chromatin in the nucleus, and consequences of their defects for genomic stability. To this end, we leverage the ease of genetic engineering and the unique tools available S. cerevisiae to investigate commonalities and specifics of mitotic versus meiotic HR. In particular we use (and keep developing) dedicated physical assays enabling step-by-step kinetics study of the pathway, which we combine with high-throughput contact genomics. The experimental data acquired are to be compared with the results of in silico simulations, in order to define the quantitative contributions of various nucleoprotein factors in the efficiency and accuracy of the repair process.
