Despite effective prevention against hepatitis B virus (HBV )infection, 300 million people worldwide are chronic HBV carriers, of whom 25% will die of liver cirrhosis or hepatocellular carcinoma (HCC). Although antiviral treatments of chronic HBV carriers, including pegylated interferon-alpha and inhibitors of the viral polymerase, control efficiently virus replication and improve liver functions, they cannot lead to a complete viral clearance. This failure is due to the inability of the treatment to eliminate the viral DNA present in the nucleus of infected cells as a covalently closed circular DNA (cccDNA). cccDNA serves as a template for the transcription of all viral RNAs and thus constitutes a viral reservoir responsible for viral rebound at the end of the treatment. Our group investigates mechanisms regulating HBV cccDNA transcription and persistence. We are studying the chromatin structure of the cccDNA and the role of cellular and viral proteins in HBV cccDNA chromatin regulation.
