Our research group has identified the Drosophila BEAF chromatin insulators (>2,000 elements) that we are currently characterizing. BEAF/CTCF insulators belong to Class I insulators that may function through common mechanisms and partners that we have biochemically identified. In particular, we test whether such factors function to regulate the expression of essential oncogenes through: a) the local organization of chromatin, nucleosome-positioning and/or the propagation of repressive chromatin/histone marks b) through long-range DNA contacts that may regulate transcription, for example by limiting enhancer-promoter interactions.
